Jodhpur is a one-fort city and that fort is the best in Rajasthan. Mehrangarh rises directly out of rock above the blue houses and the museum inside is genuinely well curated rather than a collection of dusty cases. One night is workable if you arrive early; two is better because the blue city lanes below the fort deserve a morning of their own and most people underestimate how long they will want to spend in them. The clock tower market in the evening is the other half of the city.

It works if you arrive by mid-morning and give the fort a full afternoon. Two nights is better if you want the blue city lanes and the market properly.
How far is Jaisalmer from Jodhpur?
Roughly five hours by road across open desert. It is a long, flat and quite beautiful drive, but do not plan sightseeing at either end on the same day.
